The objective of the present study was designed to establish more specific criteria for the degree of differentiation and nuclear DNA histochemical quantitation of OSCC. Morphometric measurements by image analysis computer system and DNA histochemical quantitative analysis were assessed in association with clinicopathological features of OSCC. Two histological grading systems were used; Broders’ grading system & Invasive front grading system using four histomorphological features that includes; degree of keratinization, nuclear pleomorphism, pattern of invasion and host response (inflammatory cell infiltration). The morphometric measurement was carried out using H&E staining techniques. Each nucleus and cell was submitted to morphometric parameter measurements that include nuclear area, maximum diameter (D max), form AR and D circle, cellular morphometric perimeter and nucleo: cytoplasmic ratio. A microspectrophotmerty technique was used to determine the quality of tumour nuclear DNA content with both histological grading systems of OSCC that stained by Feulgen reaction. Esterases histochemistry was employed to evaluate the distribution of carboxyl ester hydrolases in OSCC.

Random Amplified Polymorphic DNA Profiling of Bauhinia species

Different plant parts of various Bauhinia species are utilized as folk medicine. The present study was, therefore, assigned for RAPD analysis so as to explore the possibilities of applying such molecular methods to subsequent detailed biosystematic and biodiversity analysis of the Bauhinia DNAs. Bauhinia leaf samples were collected and further processed to obtain DNA. DNAs further used for PCR and RAPD analysis. RAPD primer screening was done with 15 primers and out of which, data of 9 primers were used for RAPD analysis. From distance data NJ tree was generated. The NJ tree indicates that dendrogram was divided into two groups and also showing genetic distance among plants under study. The variability exhibited at the morphological levels is also revealed upon DNA analysis of the different genotypes. The present study was done to provide foundation for advanced R&D methodologies such as the ones which employed in Biodiversity and Genetic diversity analysis. Thus it can be concluded that, RAPD analysis explore the possibilities of applying such a molecular method to subsequent detailed biosystematic and biodiversity analysis of economically important plants.

Spectrophotometric analysis of Candidal Sap in leukoplakia and OSCC

Candida species are a normal commensal which can become opportunistic under certian situations. In this study, in vitro C. albicans Sap has been evaluated and compared in nonsmokers, smokers, and patients with leukoplakia and oral squamous cell carcinoma (OSCC). Candida cell count (CCC) at 48 h was also assessed. The Sap level was measured by spectrophotometry in 38 clinical isolates of C. albicans obtained from the oral cavity of the four different groups. Sap levels and CCC were higher in individuals with leukoplakia and OSCC than in nonsmokers or smokers ; however, there was no significant difference in Sap levels or CCC between smokers and nonsmokers. Further, an intragroup correlation between CCC and Sap level was also observed. The higher level of Saps from C. albicans in individuals with leukoplakia and OSCC suggests that this pathogen plays a role in disease development and could aid in identifying the pathogenic commensal.
